The Welsh singer won best British Breakthrough Act, British Female Solo Artist and British Album for Rockferry.
But Duffy, 24, lost out to Girls Aloud and their No 1 track The Promise in the Best Single category.
Other winners included Paul Weller (British Male Solo Artist), Elbow (British Group) and Kings Of Leon (International Group).
Kanye West, 31, was named International Male Solo Artist and Katy Perry, 24, took the female equivalent; while the Pet Shop Boys received the Outstanding Contribution To Music Award.
But it was a bad night for Coldplay - despite being nominated for 4 gongs, they went home empty-handed.
Guests at the Earls Court bash included Cheryl Cole, 25, Sir Tom Jones, 68, and David Hasselhoff, 56.
James Corden, Mathew Horne and Kylie Minogue - who changed her outfit 6 times - were the presenters on the night.
